    People counting and human detection in a challenging situation

    Reliable people counting and human detection is an important problem in visual surveillance. In recent years, the field has seen many advances, but the solutions have restrictions: people must be moving, the background must be simple, and the image resolution must be high. This paper aims to develop an effective method for estimating the number of people and locate each individual in a low resolution image with complicated scenes. The contribution of this paper is threefold. First, postprocessing steps are performed on background subtraction results to estimate the number of people in a complicated scene, which includes people who are moving only slightly. Second, an Expectation Maximization (EM)-based method has been developed to locate individuals in a low resolution scene. In this method, a new cluster model is used to represent each person in the scene. The method does not require a very accurate foreground contour. Third, the number of people is used as a priori for locating individuals based on feature points. Hence, the methods for estimating the number of people and for locating individuals are connected. The developed methods have been validated based on a 4-hour video, with the number of people in the scene ranging from 36 to 222. The best result for estimating the number of people has an average error of 10% over 51 test cases. Based on the estimated number of people, some results of the EM-based method have also been shown. © 2006 IEEE.published_or_final_versio

    Human detection in crowded scenes

    In this paper, our focus is to segment the foreground area for human detection. It is assumed that the foreground region has been detected. Accurate foreground contours are not required. The developed approach adopts a modified ISM (Implicit Shape Model) to collect some typical local patches of human being and their location information. Individuals are detected by grouping some local patches in the foreground area. The method can get good results in crowded scenes. Some examples based on CAVIAR dataset have been shown. A main contribution of the paper is that ISM model and joint occlusion analysis are combined for individual segmentation. There are mainly two advantages: First, with more sufficient information inside the foreground region, even the individuals inside a dense area can also be handled. Secondly, the method does not require an accurate foreground contour.     Multicue-based crowd segmentation using appearance and motion

    In this paper, our aim is to segment a foreground region into individual persons in crowded scenes. We will focus on the combination of multiple clues for crowd segmentation. To ensure a wide range of applications, few assumptions are needed on the scenarios. In the developed method, crowd segmentation is formulated as a process to group the feature points with a human model. It is assumed that a foreground region has been detected and that an informative foreground contour is not required. The approach adopts a block-based implicit shape model (B-ISM) to collect some typical patches from a human being and assess the possibility of their occurrence in each part of a body. The combination of appearance cues with coherent motion of the feature points in each individual is considered. Some results based on the USC-Campus sequence and the CAVIAR data set have been shown. The contributions of this paper are threefold. First, a new B-ISM model is developed, and it is combined with joint occlusion analysis for crowd segmentation. The requirement for an accurate foreground contour is reduced. In addition, ambiguity in a dense area can be handled by collecting the evidences inside the crowd region based on the B-ISM. Furthermore, motion cues-which are coherent moving trajectories of feature points from individuals'are combined with appearance cues to help segment the foreground region into individuals. The usage of motion cues can be an effective supplement to appearance cues, particularly when the background is cluttered or the crowd is dense. Third, three features have been proposed to distinguish points on rigid body parts from those with articulated movements. Coherent motion of feature points on each individual can be more reliably identified by excluding points with articulated motion. © 2012 IEEE.published_or_final_versio

    Study of transforming growth factor alpha for the maintenance of human embryonic stem cells

    Human embryonic stem cells (hESCs) have great potential for regenerative medicine as they have selfregenerative and pluripotent properties. Feeder cells or their conditioned medium are required for the maintenance of hESC in the undifferentiated state. Feeder cells have been postulated to produce growth factors and extracellular molecules for maintaining hESC in culture. The present study has aimed at identifying these molecules. The gene expression of supportive feeder cells, namely human foreskin fibroblast (hFF-1) and non-supportive human lung fibroblast (WI-38) was analyzed by microarray and 445 genes were found to be differentially expressed. Gene ontology analysis showed that 20.9% and 15.5% of the products of these genes belonged to the extracellular region and regulation of transcription activity, respectively. After validation of selected differentially expressed genes in both human and mouse feeder cells, transforming growth factor a (TGFa) was chosen for functional study. The results demonstrated that knockdown or protein neutralization of TGFa in hFF-1 led to increased expression of early differentiation markers and lower attachment rates of hESC. More importantly, TGFa maintained pluripotent gene expression levels, attachment rates and pluripotency by the in vitro differentiation of H9 under non-supportive conditions. TGFa treatment activated the p44/42MAPK pathway but not the PI3K/Akt pathway. In addition, TGFa treatment increased the expression of pluripotent markers, NANOG and SSEA-3 but had no effects on the proliferation of hESCs. This study of the functional role of TGFa provides insights for the development of clinical grade hESCs for therapeutic applications. © The Author(s) 2012. © Springer-Verlag 2012.published_or_final_versio

    Heat Treated NiP–SiC Composite Coatings: Elaboration and Tribocorrosion Behaviour in NaCl Solution

    Tribocorrosion behaviour of heat-treated NiP and NiP–SiC composite coatings was investigated in a 0.6 M NaCl solution. The tribocorrosion tests were performed in a linear sliding tribometer with an electrochemical cell interface. It was analyzed the influence of SiC particles dispersion in the NiP matrix on current density developed, on coefficient of friction and on wear volume loss. The results showed that NiP–SiC composite coatings had a lower wear volume loss compared to NiP coatings. However, the incorporation of SiC particles into the metallic matrix affects the current density developed by the system during the tribocorrosion test. It was verified that not only the volume of co-deposited particles (SiC vol.%) but also the number of SiC particles per coating area unit (and consequently the SiC particles size) have made influence on the tribocorrosion behaviour of NiP–SiC composite coatings

    Interleukin-17D and Nrf2 mediate initial innate immune cell recruitment and restrict MCMV infection.

    Innate immune cells quickly infiltrate the site of pathogen entry and not only stave off infection but also initiate antigen presentation and promote adaptive immunity. The recruitment of innate leukocytes has been well studied in the context of extracellular bacterial and fungal infection but less during viral infections. We have recently shown that the understudied cytokine Interleukin (IL)-17D can mediate neutrophil, natural killer (NK) cell and monocyte infiltration in sterile inflammation and cancer. Herein, we show that early immune cell accumulation at the peritoneal site of infection by mouse cytomegalovirus (MCMV) is mediated by IL-17D. Mice deficient in IL-17D or the transcription factor Nuclear factor (erythroid-derived 2)-like 2 (Nrf2), an inducer of IL-17D, featured an early decreased number of innate immune cells at the point of viral entry and were more susceptible to MCMV infection. Interestingly, we were able to artificially induce innate leukocyte infiltration by applying the Nrf2 activator tert-butylhydroquinone (tBHQ), which rendered mice less susceptible to MCMV infection. Our results implicate the Nrf2/IL-17D axis as a sensor of viral infection and suggest therapeutic benefit in boosting this pathway to promote innate antiviral responses

    Is telomere length socially patterned? Evidence from the West of Scotland Twenty-07 study

    Lower socioeconomic status (SES) is strongly associated with an increased risk of morbidity and premature mortality, but it is not known if the same is true for telomere length, a marker often used to assess biological ageing. The West of Scotland Twenty-07 Study was used to investigate this and consists of three cohorts aged approximately 35 (N = 775), 55 (N = 866) and 75 years (N = 544) at the time of telomere length measurement. Four sets of measurements of SES were investigated: those collected contemporaneously with telomere length assessment, educational markers, SES in childhood and SES over the preceding twenty years. We found mixed evidence for an association between SES and telomere length. In 35-year-olds, many of the education and childhood SES measures were associated with telomere length, i.e. those in poorer circumstances had shorter telomeres, as was intergenerational social mobility, but not accumulated disadvantage. A crude estimate showed that, at the same chronological age, social renters, for example, were nine years (biologically) older than home owners. No consistent associations were apparent in those aged 55 or 75. There is evidence of an association between SES and telomere length, but only in younger adults and most strongly using education and childhood SES measures. These results may reflect that childhood is a sensitive period for telomere attrition. The cohort differences are possibly the result of survival bias suppressing the SES-telomere association; cohort effects with regard different experiences of SES; or telomere possibly being a less effective marker of biological ageing at older ages

    Protein Profile Changes during Porcine Oocyte Aging and Effects of Caffeine on Protein Expression Patterns

    It has been shown that oocyte aging critically affects reproduction and development. By using proteomic tools, in the present study, changes in protein profiles during porcine oocyte aging and effects of caffeine on oocyte aging were investigated. By comparing control MII oocytes with aging MII oocytes, we identified 23 proteins that were up-regulated and 3 proteins that were down-regulated during the aging process. In caffeine-treated oocytes, 6 proteins were identified as up-regulated and 12 proteins were identified as down-regulated. A total of 38 differentially expressed proteins grouped into 5 regulation patterns were determined to relate to the aging and anti-aging process. By using the Gene Ontology system, we found that numerous functional gene products involved in metabolism, stress response, reactive oxygen species and cell cycle regulation were differentially expressed during the oocyte aging process, and most of these proteins are for the first time reported in our study, including 2 novel proteins. In addition, several proteins were found to be modified during oocyte aging. These data contribute new information that may be useful for future research on cellular aging and for improvement of oocyte quality

    Room temperature continuous–wave green lasing from an InGaN microdisk on silicon

    Optically pumped green lasing with an ultra low threshold has been achieved using an InGaN/GaN based micro-disk with an undercut structure on silicon substrates. The micro-disks with a diameter of around 1 μm were fabricated by means of a combination of a cost-effective silica micro-sphere approach, dry-etching and subsequent chemical etching. The combination of these techniques both minimises the roughness of the sidewalls of the micro-disks and also produces excellent circular geometry. Utilizing this fabrication process, lasing has been achieved at room temperature under optical pumping from a continuous-wave laser diode. The threshold for lasing is as low as 1 kW/cm2. Time–resolved micro photoluminescence (PL) and confocal PL measurements have been performed in order to further confirm the lasing action in whispering gallery modes and also investigate the excitonic recombination dynamics of the lasing
